I would suggest taking the NYS Thruway I-90 west to the Geneva exit (42) then going down the west side of Seneca Lake (rte 14) to Watkins Glen. From there, you can take 414/227/79 to Ithaca which is at the southern end of Cayuga Lake. From Ithaca you can travel up the west side of Cayuga Lake and hit the wineries there along Rte 89. At the top of Cayuga you head back west a little and get back on the Thruway at exit 41 (Seneca Falls). This way, you do a nice loop where you get to see two of the biggest and nicest Finger Lakes and a lot of really good wineries. Here's a link to a site with some maps.
